Release mechanism and rate can be tuned by changing polymer concentration, size and shape of nanofibers. In the present study, a drug delivery system was prepared by natural polymers of gum tragacanth and gelatin using electrospinning method for control delivery of imipenem/cilastatin in antibacterial applications.Methods: Nanofibers' diameters and morphology were measured by scanning electron microscopy (SEM). The physicochemical properties of nanofibers' materials were analyzed by Fourier transform infrared spectroscopy (FTIR). In vitro release of drug was investigated using ultraviolet (UV) spectroscopy. MTT assay was performed using L929 (NCBI C161) cell line by extraction method on nanofibers with/without drug. The antibacterial activity was performed on nanofibers against Escherichia coli and Staphylococcus aureusbacteria. Findings: Nanofibers with concentration ratio of gum tragacanth/gelatin (80 to 20 %) were selected for further investigation. Diameters of nanofiber with/without drug were in the range of 100 and 200 nm, respectively. Also, the results confirmed that there is no possible interaction between drug and nanofiber materials. The drug release profile from nanofibers showed 82% release in 120 h. The result of MTT assay indicated that nanofibers without drug had no toxicity effects. Also cell viability of drug-loaded nanofibers has reached about 91% cell viability after 5 days. Overall, this study confirmed that this nanofiber could potentially be used as a drug carrier for antibacterial agent delivery, specifically against gram-negative bacteria without cytotoxicity effect.}, keywords = {nanofibers,gum tragacanth,gelatin,imipenem/cilastatin,drug delivery system}, title_fa = {تهیه و بررسی نانوالیاف کتیرا-ژلاتین در سامانه‌های دارورسانی ضدباکتریایی}, abstract_fa = {فرضیه: از میان سامانه‌های مختلف دارورسانی، نانوالیاف پلیمری به‌عنوان حامل مناسب دارو در مطالعات گذشته استفاده شده‌اند. سرعت و سازوکار رهایش دارو از نانوالیاف پلیمری را می‌توان با تغییر غلظت پلیمر، اندازه و شکل‌ نانوالیاف کنترل کرد. در پژوهش حاضر، سامانه دارورسانی با استفاده از پلیمرهای طبیعی کتیرا و ژلاتین برای رهایش کنترل‌شده داروی ایمی‌پنم-سیلاستاتین در کاربردهای ضدباکتریایی تهیه شد.روش‌ها: قطر و شکل‌شناسی نانوالیاف با میکروسکوپی الکترونی پویشی (SEM) و خواص فیزیکی شیمیایی نانوالیاف با طیف‌سنجی زیرقرمز تبدیل فوریه (FTIR) بررسی و تحلیل شد. رهایش برون‌تنی داروی ایمی‌پنم-سیلاستاتین با طیف‌سنجی فرابنفش (UV) بررسی شد. آزمون سمیت سلولی (MTT) با استفاده از سلول (NCBI C161)و L929 و روش استخراج روی نانوالیاف با دارو و بدون آن انجام شد. فعالیت ضدباکتریایی نانوالیاف در برابر باکتری‌های اشرشیا کلی و استافیلوکوکوس اورئوس مطالعه شد.یافته‌ها:  نانوالیاف با نسبت 80 به 20 درصد کتیرا به ژلاتین برای بررسی بیشتر انتخاب شدند. نتایج  نشان داد، قطر نانوالیاف با دارو و بدون آن به ترتیب در محدوده 100 و nm 200 بوده و دارو با نانوالیاف هیچ بر‌هم‌کنش شیمیایی نداشته است. نیم‌رخ رهایش دارو از نانوالیاف، آزادسازی %82 دارو را طی h 120 نشان داد. نتایج آزمون سمیت سلولی نشان داد، نانوالیاف بدون دارو هیچ اثر سمی نداشته‌اند. زنده‌مانی سلول‌ در نانوالیاف بارگذاری‌شده با دارو نیز پس از 5 روز حدود %91 بوده است. به‌طور کلی این مطالعه تأیید کرد، نانوالیاف تهیه‌شده می‌توانند به‌عنوان حامل مناسبی برای رساندن عامل ضدباکتریایی، به‌ویژه در برابر باکتری‌های گرم منفی بدون اثر سمیت سلولی استفاده شوند.}, keywords_fa = {نانوالیاف,کتیرا,ژلاتین,ایمی‌پنم-سیلاستاتین,سامانه دارورسانی}, url = {http://jips.ippi.ac.ir/article_1650.html}, eprint = {http://jips.ippi.ac.ir/article_1650_1c09f3917f985dcc02084879fe2f90f1.pdf} }
